I have a 2012 Defy1 and a 2015 Defy2 and I don't recall either being oddly spaced.
As mentioned previously, I also have mudguards on the Defy1 and as they sit so close to the wheel even if you could get the tail end to fit by tweaking the stays it wouldn't fit properly under the brake and where the mudguard attaches to the seat tube.
You can use these points as reference on yours.
Does the centre of the tyre align with the bolt above the derrailleur on the seat tube
Does the centre of the tyre align with the centre of the brake caliper bolt
