Ive had a couple of punctures recently and whilst have replcaed the tubes have attempted to repair the puncture with the pre gluded patches. However leaving the tube inflated overnight, with no obvious air escaping, needless to say the next moring it is flat. Any ideas where I'm going wrong?

Not certain, but if repairing a tube at home I use regular patches and rubber solution, on the road I use a spare tube. I don't know what preglued patches are like now but I used to regard them a a bit of a temporary solution- pressure sensitive type adhesives just don't work so well as rubber solution (although they can work well-enough).

When you say leaving tube inflated overnight I take it that you mean inflated and in the tyre? preglued patches don't stick well enough to seal a tube without having the tyre in place.

You may be doing nothing wrong. Remember when a tube is inflated inside a tyre, the pressure pushing the tube and patch against the inside of the tyre helps make the seal. Testing a tube with more than a few psi outside of a tyre can often fail.
That said, have you checked for leaks from the valve? Sheider valves have a screw in core that can become loose and Presta can also leak through the valve if the innards don't reseal correctly.
Put a few psi in the tyre that's failed (before you peel the failed patch off) just to confirm where the leak is coming from.
Finally - are you sure the tube didn't have more than one puncture?

If using pre glued patches you still need to prepare the tyre the same as ordinary patches.Roughen up the area around the leak, apply patch and press firmly for a couple of minutes. Put tube in tyre and inflate. So far I haven't had a problem using this method.

This.^^^...it's all in the preparation.
And do not inflate a repaired tube outside of a tyre.
