Not shifting

I have just changed my freehub body(9 speed), put a 10 speed cassette on,and chain, all new. I can shift up one cog but then nothing, will not even shift back down! The deurailleur pushes up by hand, but no shifting I have tried the barrel adjustment to no avail. Any ideas please?? (Road bike)

I managed to adjust and setup from scratch the GF's bikes gears using the info there, and all seems well.
