is this to do with cable tension ? It wasn't going onto smallest teeth at back so basically I twiddled barrel adjuster and messed about with limit screws and now it is engaging , but I rode bike today and now it won't go "up " the cassette ( 12^14^16 and so on ) so have I turned barrel adjuster too much

Probably. Start again and follow some instructions on how to set up indexing. Limit screws only set the limits: they'll only come into play if gears are inaccessible or the chain is falling off. The most important thing is cable tension on the two smallest cogs IMO, which you don't mention touching. The barrel adjuster is the second but mainly for small adjustments. Keeping the cable clean and lubed is third.

If shifting is slow or not spot on, turn the barrel adjuster towards the way you want it to change. Turn towards the bike if not changing up, turn away from the bike if not changing down.
