I have a Defy 2 2011 with a 34/50 crankset and 12/25 cassette. I want a lower low gear and am thinking of changing the cassette to a 12/28. Is this the best/easiest/cheapest route? Will the chain need to be changed? anything else?

I think you probably just need to change the cassette and nowt else. My wife had a 12/27 cassette and I swapped it for a 12/30 (only available in Tiagra) - didn't need to change the chain.

I have the Defy 2 2010.

The 28 will fit but is on the limit and may need the rear mech tweeking.

Fit a 12-27 rear and it will fit straight on with no adjustments.

(indexing might require a very slight tweek, mine didnt.

I am taking it as the 9 speed tiagra

It's the 9 speed Sora (Tigra was on the 2010 and 2012 Defy 2). 12-27 sounds simpler then...

You'll also need a longer chain.

Yip thats the easiest/cheapest way. You'll need a longer chain too, you might be able to lengthen your older chain with quicklinks but a new one will marry up with your new cassette better. Only if you go larger than 28t will you need a new derailleur too if its a short cage.

I still have a few links left of the chain I recently fitted. Will a 12-27 be simpler as posted above? I don't want to replace the derailleur.

12-27 will be fine.

I didnt need to lengthen the chain, although big ring to large rear is at the limit its still has some movement in the mech.

If you want to go to 30 or 32 then go for a deore reaer mech, cassette and new chain

I believe so, a 11-26 certainly is no fiddling needed (my experience). I've lengthened a relatively new chain with quick links but if your chain is not new for the price of a new one you'll be just as well to buy new and you won't have the potential problem of different worn chains wearing out a new cassette and causing slippage.

I did changing from an 12-25 to an 11-26, it depends how the original was set up.

I can get an ultegra 12-27 for about £45 or a sora/tiagra HG50 12-27 for about £22. I know the Ultegra is going to be better quality and a bit lighter but is it really twice as good? What to do?
