HELP- Shimano backwards compatibility

I'm trying to get an ultega 6500 shifter...

to play nice with a 105 5600 rear mec... (9 and 10 spd compatible)

but i'm having some problems, should this work though in theory? I have a varsity hill climb to do this afternoon, I should not have messed with my setup this morning! but please help

If the number of sprockets matches the number of clicks in the RH lever it should be plain sailing. What's the troublem?

+1. The standard 6500 assumes the rear wheel has a Shimano compatible 9 speed cassette.
